Evidence for the utilization of host tRNA(m 5 U)methylase to modify tRNA coded by phage T4.

چکیده
Mutants of Escherichia coli defective only in the biosynthesis of 5-methyluridine (ribothymidine) in their transfer ribonucleic acid (tRNA) were employed to investigate whether phage T4 induces its own tRNA(mV)methylase and whether T4-specific tRNA contains 5-methyluridine. The amounts of other methylated derivatives in the T4-specific tRNA were also determined. The results establish that T4-specific tRNA contains 5-methyluridine, and that there is an absolute requirement for a functional host tRNA(mW)methylase to undertake this modification. Comparison of several physical properties of the host enzyme before and after phage T4 infection did not suggest any phage-directed alteration of the enzyme. The distribution of the methylated constituents in T4-specific tRNA is distinguishable from that in host tRNA. This change, however, may simply reflect a different population of tRNA chains produced by the T4 phage, rather than some change in the tRNA methylase activity of the host: _ -.

Valyl-tRNA synthetase modification-dependent restriction of bacteriophage T4.
A strain of Escherichia coli, CP 790302, severely restricts the growth of wild-type bacteriophage T4. In broth culture, most infections of single cells are abortive, although a few infected cells exhibit reduced burst sizes. In contrast, bacteriophage T4 mutants impaired in the ability to modify valyl-tRNA synthetase develop normally on this strain. Biochemical evidence indicates that the phage...

T4 mutants lacking polynucleotide kinase (pnk-) or RNA ligase (rli-) do not grow on E. coli CTr5x. During the abortive infections there accumulate host tRNA fragments that match into two species severed 3' to the anticodon. The CTr5x-specific fragments appear only transiently with wt phage, implicating the affected enzymes in phosphoryl group rearrangement and religation [David et al. (1982) Vi...
متن کاملEffect of T4 modification of host valyl-tRNA synthetase on enzyme action in vivo.
It has previously been found that bacteriophage T4 modifies the valyl-tRNA synthetase of its host; in this work we have examined the effects of this modification on the action of the enzyme in uiuo. For this purpose we compared the properties of T4-infected cells with those of uninfected cells or cells infected with the amber mutant 0~2, which by all available criteria seems to leave the host e...
متن کاملResponse of a phage modification factor to enhanced production of its target molecule.
Escherichia coli cells with plasmids bearing the valS gene were constructed from the Clark-Carbon collection. Their increased valyl-tRNA synthetase (EC 6.1.1.9) activity was not matched by commensurate production of a modifying peptide during T4 infection. Thus, phage vs gene expression is set to modify the normal amount of valyl-tRNA synthetase activity.
متن کاملPurification and properties of a T4 bacteriophage factor that modifies valyl-tRNA synthetase of Escherichia coli.
After T4 bacteriophage infects Escherichia coli, a peptide tau, produced under the control of a phage gene, binds to the host valyl transfer ribonucleic acid synthetase (EC 6.1.1.9) and thereby changes several of its physicochemical properties. The interaction of tau with the host enzyme was investigated in vitro after extensively purifying the factor from T4-infected E. coli using a rapid puri...
